Durham originated in 1853 to satisfy the need for a suitable railroad depot for the North Carolina Railroad between Wilson and Hillsborough. The landowners at nearby Prattsburg refused to sell land to the railroad, but a country physician named Bartlett Durham who lived and practiced medicine a little further to the northwest, donated land to the railroad. Subsequently, the depot was named Durham Station as a token of appreciation for his generosity.
